When the two participants have different biological mothers, the X chromosome analysis is more straightforward, as it is not necessary to separate the maternal component.

The study is based on the analysis of STR genetic markers of the X chromosome. Since both women have inherited an X chromosome from their father, it is possible to directly compare this genetic component between them.

Since they do not share a mother, any relevant coincidence in the X chromosome analyzed is attributable to paternal origin. Therefore, the genetic profiles of both participants are compared to assess for compatibility.

If the profiles match, the result is compatible with both sharing the same biological father. On the contrary, if they do not match, a paternal relationship can be excluded.

In the case where the two participants share the same biological mother, it is necessary to also analyze the maternal DNA in order to correctly study the paternal relationship.

The analysis is based on the study of STR genetic markers of the X chromosome. Since each daughter inherits one X chromosome from the mother and one from the father, it is essential to identify and separate the maternal component. For this purpose, the mother’s DNA is analyzed, which makes it possible to determine which part of each daughter’s genetic profile comes from her.

Once this maternal component is isolated, the X chromosome of paternal origin can be identified in each participant. These chromosomes are compared with each other to assess whether there is a genetic match.

If the profiles match, the result is compatible with both sharing the same biological father. If they do not match, a paternal relationship can be excluded.
